Selby, renowned for his high strike rate in close matches, fought from 3-1 behind and needed just one chance in the decider to clinch a terrific match.
He now faces Mark King for a place in the final, after he defeated Anthony Hamilton 5-2 in what turned out to be a controversial match.
Hamilton, who beat Ronnie O'Sullivan yesterday blamed the loss on the cameraman - "The cameraman was moving on every shot, all the way through the match," said a furious Hamilton.
Murphy seemed to have control of his quarter-final against Selby, leading 3-1 at the interval courtesy of breaks of 70 and 85.
However Wuxi Classic winner Selby fought back in style, winning frame five with a 70, before Murphy restored the lead at 4-2.
Selby then levelled up at 4-4, taking the match into a decider, something which Murphy had experienced twice already in the tournament.
However this time he came unstuck, failing to find safety out of a Snooker and allowing Selby to clean up in superb fashion with a 117 for victory.
World number one Mark Williams continued his fine form, whitewashing a sorry Matthew Stevens 5-0 to set up an unexpected semi-final clash with Neil Robertson, after the Aussie dumped out world champion John Higgins 5-2 with a fine display of potting.
Semi-final action begins tomorrow at 7am UK time, with both matches streamed live over the best of 11 frames.
