The 24 year-old Leicester cueist had never previously made it past the last 32 of a major event and came into the PTC tournament as a rank outsider.
He won the first frame of the final with a 70 break, before Dott secured a snooker on the last red and preceded to clear for 1-1.
Dott, who went into the final as overwhelming favourite having already beaten John Higgins, Stephen Hendry and Ronnie O'Sullivan to get there, won a tight third frame to lead 2-1.
Woollaston then turned on the style, dominating the last three frames, with top runs of 42, 43 and 50 enough to seal the title and £10,000 first prize.
He joins Ronnie O'Sullivan and Judd Trump as a winner of a PTC event this season, also bolstering his chances of qualifying for a TV major with the acquired 2,000 ranking points.
"Hopefully I am starting to fulfil my potential now," said Woollaston. "I'm guaranteed to be in the PTC finals now and my target for the season is to get into the top 48 and qualify for a few venues."
"I didn't play a bad match in the tournament, the only time I was worried was in the quarter-finals when I was 2-0 down to Alan McManus. I came back to win that 4-3, and in the final against Graeme I did well to recover from 2-1 down," he added.
The PTC tour moves overseas to Germany next week for PTC 4, The Paul Hunter Classic.
